Sirtuins & NAD+

Longevity science · How this pathway drives aging · How Midas addresses it

Sirtuins are a family of seven NAD+-dependent deacylase enzymes (SIRT1-SIRT7) that regulate virtually every aspect of cellular health: DNA repair, gene silencing, mitochondrial biogenesis, inflammation, circadian rhythm, and stress response. They are often called "longevity enzymes" because their activation extends lifespan in model organisms. The critical bottleneck is NAD+. Every sirtuin reaction consumes one molecule of NAD+, and NAD+ levels decline ~50% between ages 40-60. As NAD+ falls, sirtuin activity drops, and the protective processes they govern deteriorate. The Midas Protocol includes NAD+ 500mg specifically to restore substrate availability for all seven sirtuins. Combined with Epitalon (which SIRT1 helps maintain telomeres) and FOXO4-DRI (which removes cells that consume NAD+ via CD38 upregulation), the Protocol addresses both NAD+ supply and upstream causes of its decline.

The Midas Approach

The Midas Longevity Protocol doesn't address sirtuins & nad+ in isolation — it targets this pathway alongside 9 other aging mechanisms simultaneously. This multi-pathway approach reflects the scientific consensus that aging is not a single process but a network of interconnected pathways (Lopez-Otin et al., Cell 2023, "Hallmarks of Aging").
